近日,來自湖南大學化學生物傳感與計量學國家重點實驗室的聶舟教授課題組在細胞調控領域取得重要進展,在國際頂級期刊《Angew. Chem. Int. Ed.》(IF=12.102)在線發表研究論文《A DNA-mediated Chemically Induced Dimerization (D-CID) Nanodevice for Nongenetic Engineering of Receptor to Control Cell Behavior》,并被該期刊選為“Hot Paper”。

細胞行為的精確控制是目前細胞治療技術中的難點和熱點問題。
聶舟教授課題組開發了一種基于DNA納米機器的受體非遺傳改造策略,對細胞膜表面受體特異性進行定制化改造,通過DNA納米技術賦予其化學小分子響應性,實現了小分子調控的細胞行為。 該策略應用DNA介導化學誘導二聚(D-CID)實現了受體酪氨酸激酶的二聚激活,并介導下游的細胞信號傳導途徑,可對細胞遷移行為進行精確控制,無需復雜遺傳改造即可賦予細胞特定的化學趨向性。進一步通過不同的DNA功能模塊組合,并利用微流控裝置構建復雜多細胞環境,成功證明了該策略可用于多目標細胞行為的多化學因子精準正交控制,有望實現在復雜細胞環境下對特定靶細胞類型的智能行為調控。 D-CID策略由于其DNA識別單元的多樣性,因此具有小分子配體及對應細胞膜受體的普適性。 這種細胞行為的智能化調控策略有望為細胞治療及再生醫學的體內應用提供一種新思路。 使用產品
